DIM stands for di-indolyl-methane. DIM comes from indole-3-carbinol, which is a compound naturally found in cruciferous vegetables such as broccoli, cauliflower, Brussels sprouts, cabbage, and collards. When these foods reach the stomach, indole-3-carbinol is broken down by the stomach acid. DIM is then released as a by-product.

However, the amount of DIM you get from digesting a serving or two of broccoli isnt going to be enough if youre in a state of estrogen overload. Youd need to eat about 6 pounds of these vegetables a day to get a therapeutic dose of DIM.

Thats where supplements come in. A clinical trial published in the journal Nutrition and Cancer has reported that it takes a dosage of about 100 mg of DIM to have any effect on estrogen dominance.

While taking indole-3-carbinol can be helpful for some women, I do not recommend taking supplemental I3C for two reasons:

The conclusion is: DIM works a lot better, and it does so without the potential negative effects you can get from taking I3C. This molecule is so great for getting rid of excess estrogen that its often referred to as the Estrogen Buster. However, its not a complete story.

What Are Some Of Dims Opposite Effects On Breast Cancer Cells

I3C studies show that I3C stops the growth of estrogen receptor-positive and negative breast cancer cells in culture.10 DIM reportedly either makes estrogen receptor positive breast cancer cells grow11 or inhibits them9 in culture. According to researchers at the University of California, DIM promotes the growth of human breast cancer cells about as half as well as estrogen when no estrogen is present in the culture. In addition, DIMs inhibition of cancer cell growth was weak in the presence of estrogen. Remember, these are laboratory conditions only and probably dont occur in real life. How DIM behaves, however, may depend on dose. Researchers at Texas A& M report that DIM significantly counteracts estrogen-induced growth of MCF-7 cells in culture at higher doses. However, unlike I3C which retards the growth of estrogen receptor negative breast cancer cells, DIM has no effect on estrogen receptor negative cells.10 Another question about DIM is whether it can increase aromatase in breast and other tissue. Aromatase is an enzyme that helps create estrogen. DIM reportedly enhances the enzyme in adrenocortical cancer cells.11

Not only is DIM not responsible for the anti-cancer effects of I3C, it has fewer anti-cancer effects , when its isolated from the other phytochemicals that naturally occur with it.

Steroid Hormone Concentrations In Serum Samples And Tumor Homogenates

At the time of necropsy of the control and experimental SCID mice, 1 ml of blood was obtained by cardiac puncture using a 25G needle from each mouse. Blood samples were collected in special tubes and spun down in a refrigerated centrifuge at 4 °C and a speed of 1200 x g for 20 min. Serum was harvested and stored at 20 °C until assayed for the presence of hormones. Frozen tissue specimens of approximately 0.10.5 g from individual mice were homogenized in 5 ml of PBS and the homogenate centrifuged at 1200 x g, for 20 min at 4 °C. The supernatants were harvested and stored at 80 °C until hormone assays .

Levels of estrone sulphate , estradiol , androstenedione , testosterone , dehydroepiandrosterone and progesterone in tumor homogenates and serum samples were assayed by amplified Enzyme-Immunoassay previously validated in our laboratory .

Cell Culture Vectors And Indole

The cell lines used in this study were obtained from the American Type Culture Collection and maintained as monolayer cultures in Dulbecco’s modified Eagle’s medium supplemented with 10% fetal calf serum , 2 mmol/L glutamine, 0.1 mg/L streptomycin and 1000 U/L penicillin G . All of the vectors used in this study were described previously , including the ER expression plasmid containing both the T7 and cytomegalovirus promoters, the ERE-TK-LUC reporter composed of the vitellogenin A2 ERE controlling a minimal thymidine kinase promoter , and luciferase in plasmid pGL2, the expression of wild-type BRCA1 in pcDNA3 plasmid, pCMV-Sp1 and Sp1-TK-LUC. I3C was purchased from Sigma , dissolved in 95% ethanol and stored at 20°C before use. The MTT assay of I3C toxicity showed that IC3 resulted in a significant toxicity in these cell lines . Therefore, 10125 mol/L doses of I3C were employed in the present study.

The protocol was approved by the Committee of the Universidad Complutense of Madrid, and Animal Protection Area of the Community of Madrid, Spain . Twenty-four non-ovariectomized female SCID mice , 68 weeks of age and weighing between 20 and 22 g were used. The animals were housed in a flexible-film isolator in cages , each measuring 330 cm2×12 cm, in a room with controlled environmental conditions .
